چکیده

Linear poly(VPGVG) has been extensively studied over the years as a model for the structural protein elastin. Elastin is characterized by a lower critical solution temperature (LCST). This LCST can be influenced by several factors, mainly molecular weight, concentration, and pH. An ABA type block copolymer was synthesized using atom transfer radical polymerization (ATRP), in which the VPGVG sequence is in the side chain of the A block and the B block is poly(ethylene glycol) (PEG). The LCST behavior of this series of elastin based side chain block copolymers was investigated by changing the degree of polymerization, polymer concentration, and pH. The effects of these parameters on the LCST of the side chain block copolymers were similar when compared with linear poly(VPGVG). The aggregates which were formed above the transition temperature were investigated using light scattering and cryo-SEM techniques.

Fine-tuning the transition temperature of a stimuli-responsive polymer by a simple blending procedure.

Characterization of the 4D5Flu single-chain antibody with a stimulus-responsive elastin-like peptide linker: a potential reporter of peptide linker conformation.

Single-chain antibodies (scFvs) are comprised of IgG variable light and variable heavy domains tethered together by a peptide linker whose length and sequence can affect antigen binding properties. The ability to modulate antigen binding affinity through the use of environmental triggers would be of great interest for many biotechnological applications. We have characterized the antigen binding...

Force Spectroscopy of Stimulus-Responsive Polymers

Interactions on the level of single macromolecules become exceedingly important as we strive to understand and harness the properties of nanoscale systems. Specifically, we are interested in the nanomechanical behavior of stimulus-responsive polymers that display a strong volume transition which can be exploited for force generation. For example, the genetically encodable synthesis of stimulus ...

Quantitative model of the phase behavior of recombinant pH-responsive elastin-like polypeptides.

Quantitative models are required to engineer biomaterials with environmentally responsive properties. With this goal in mind, we developed a model that describes the pH-dependent phase behavior of a class of stimulus responsive elastin-like polypeptides (ELPs) that undergo reversible phase separation in response to their solution environment. Under isothermal conditions, charged ELPs can underg...

Bioinspired Poly(2-oxazoline)s

Poly(2-oxazoline)s are regarded as pseudopeptides, thus bioinspired polymers, due to their structural relationship to polypeptides. Materials and solution properties can be tuned by varying the side-chain (hydrophilic-hydrophobic, chiral, bioorganic, etc.), opening the way to advanced stimulus-responsive materials and complex colloidal structures. The bioinspired ―smart‖ solution and aggregatio...
